Differentiation relies on Glucose fermentation, which produces acid and a characteristic red color change in colonies (inferred from the Neutral Red dye in the base formula). The overlay procedure, standard for use with this agar, suppresses non-fermentative organisms, ensuring a highly accurate Enterobacteriaceae count.
