LabChem’s Carbol-Fuchsin Solution (Kinyoun) is a specialized primary stain designed for the rapid and safe identification of acid-fast bacteria, such as Mycobacterium tuberculosis and Nocardia. As a modification of the traditional Ziehl-Neelsen stain, the Kinyoun formulation utilizes a higher concentration of phenol and basic fuchsin, allowing the dye to penetrate the waxy, mycolic acid-rich cell walls of these organisms without the application of heat. This "cold method" significantly improves laboratory safety by eliminating the hazards of heating phenolic solutions. The result is a vibrant, bright red/pink staining of acid-fast bacilli (AFB) that resists decolorization by acid-alcohol, providing a high-contrast visual against a methylene blue or malachite green counterstain.
The solution’s primary benefit is its precise formulation, which allows for staining without the application of heat (the cold method), improving safety and workflow convenience. Carbol-Fuchsin is a powerful basic dye that binds strongly to the high-mycolic acid content of the bacterial cell wall, which resists decolorization by acid-alcohol, resulting in the characteristic bright red/pink staining of acid-fast bacilli. Choosing this ready-to-use formulation eliminates hazardous and time-consuming in-house preparation, guaranteeing consistent staining intensity every time.
Features:
Enhanced Safety: By removing the requirement for Bunsen burners or hot plates during the staining process, the Kinyoun method reduces the inhalation of phenolic vapors and prevents accidental fire hazards.
Workflow Efficiency: Pre-mixed and ready to use, this solution saves valuable technician time and guarantees lot-to-lot consistency for clinical and research diagnostics.
Superior Penetration: The high phenol content acts as a powerful solvent, ensuring the basic fuchsin dye binds strongly to the lipid-rich cell walls of mycobacteria.
Reliable Diagnostics: Specifically formulated to meet the rigorous requirements of tuberculosis screening and environmental microbiology.
Laboratory Protocol & Safety:
Chemical Hazard: This product contains phenol and basic fuchsin. It is known to the State of California to cause cancer and reproductive harm (Prop 65). Always handle in a well-ventilated area or fume hood using appropriate PPE (gloves, goggles, and lab coat).
Decolorization: After staining with Carbol-Fuchsin, ensure the slide is treated with Acid-Alcohol until the run-off is clear. Only acid-fast organisms will retain the primary pink stain.
Storage: Store at room temperature (15°C to 30°C) and keep the bottle tightly closed. Protect from excessive light to prevent the degradation of the basic fuchsin dye.
